New nano/microfluidic platforms for biosensing applications

Abstract: Micro/nanofluidics has emerged in recent years as a powerful method of manipulating fluids at small length-scales. To highlight their versatility and great potentials, I present 2 simple nano/microfluidic platforms to showcase how nano- and microfabrication techniques can be coupled with fluidics for nanoplasmonic biosensing and immunoassay applications.


(a) Nanoplasmonic platforms for multiple biosensing applications: We develop a nanoplasmonic platform with remarkable biocompatibility, for long-term survival of prokaryotic and eukaryotic cells, thereby enabling nanoplasmonic based cell bioassays. This nanoplasmonic platform is used for monitoring mitosis of fibroblasts for 7 days, E. coli biofilm formation, and DNA polymerase activity in real-time. In addition, the same nanoplasmonic platform is used to conduct routine protein/DNA based immunoassays with a limit of detection of ∼66 zM.


(b) Making Waves in a Total Capture Nanofluidic Immunoassay: understanding nanoconfinement phenomena is necessary to develop nanofluidic technology platforms. We demonstrate a convection limited nanofluidic immunoassay that achieves total capture of a target analyte, and an apparent shift in the reaction equilibrium due to nanoconfinement.
