Short Abstract: This thesis is dedicated to advancing aptamer-based diagnostic technologies, with a specific focus on developing point-of-care (PoC) devices designed to detect small molecule biomarkers. By exploring aptamer design, conjugation methods, and SELEX processes, the research aims to enhance the detection and stability of diagnostic platforms. The study delves into various assay formats and assesses aptamer binding affinity, specificity, and sensitivity for small molecule biomarkers, particularly optimizing lateral flow assays (LFAs). Key factors such as nucleic acid immobilization, blocking agents, and chemical modifications are thoroughly analyzed to improve assay performance.

Additionally, this work evaluates different DNA-gold nanoparticle (DNA-AuNP) conjugation techniques and storage conditions to enhance the stability and functionality of nanoparticles used in LFAs. The research also introduces innovative SELEX methodologies for aptamer selection, integrating next-generation sequencing (NGS) to streamline the discovery process. These endeavors collectively contribute to developing more reliable, sensitive, and specific PoC diagnostic tools for small molecule biomarkers, laying a robust foundation for future applications in medical diagnostics.
